Abstract

The synergistic combination of gold nanoparticles (AuNPs) and collagen in advanced skincare formulations offers a promising approach for cutaneous rejuvenation and wound repair. This review examines the complementary mechanisms by which AuNPs enhance collagen stability and bioavailability while promoting fibroblast activity and extracellular matrix remodeling. We analyze current evidence demonstrating that AuNPs (5-50 nm) can: (1) protect endogenous collagen from UV-induced degradation through antioxidant effects, (2) stimulate de novo collagen synthesis via TGF-β pathway activation, and (3) improve the thermal stability and transdermal delivery of topical collagen. Clinical studies show formulations combining 10-20 ppm AuNPs with hydrolyzed collagen peptides achieve 28-42% improvement in skin elasticity parameters compared to collagen-only preparations. However, we critically address formulation challenges including nanoparticle aggregation in collagen matrices and long-term safety considerations. Emerging strategies such as collagen-capped AuNPs and light-activated systems demonstrate enhanced targeting of photoaged skin while minimizing potential cytotoxicity. This work provides a framework for optimizing AuNP-collagen synergies in next-generation cosmeceuticals and therapeutic dermatology.
