Objective: To study NRP-1 gene expression in nasopharyngeal carcinoma tissue and the regulating effect of targeted inhibition of gene expression on cell function. Methods: Nasopharyngeal carcinoma tissue, adjacent tissue and normal tissue were collected, and NRP-1 expression levels were detected; nasopharyngeal carcinoma CNE-2 cells were cultured and transfected with NRP-1 siRNA and negative control (NC) siRNA, and the contents of angiogenesis molecules, proliferation-related proteins and invasion-related proteins were detected. Results: NRP-1 protein content, positive cell rate, staining intensity and IHC score in nasopharyngeal carcinoma tissue were higher than those in adjacent tissue and normal tissue; VEGF, bFGF, EGF and ANG-1 contents in culture medium of NRP1-siRNA group were lower than those in NC-siRNA group; Tie-2, EGFR, Livin, Bmi-1, CDK6, CDK4, CCND1, E2F1, LMP1, FN1, MT1-MMP and Fascin expression levels in cells of NRP1-siRNA group were lower than those in NC-siRNA group. Conclusions: NRP-1 expression level significantly increases in nasopharyngeal carcinoma tissue, and NRP-1 can promote nasopharyngeal carcinoma angiogenesis, cell proliferation and invasion.
